AstraZeneca Prostate Cancer Drug Receives Expanded FDA Approval
The U.S. Food and Drug Administration (FDA) has broadened the approved uses for AstraZeneca's drug, Truqap (capivasertib), in treating prostate cancer. This expansion allows for its application in a wider patient population previously not covered. This is the first new drug for this use in a decade.

The pharmaceutical company AstraZeneca has secured an expanded approval from the U.S. Food and Drug Administration (FDA) for its medication, Truqap, also known by its generic name capivasertib. This updated clearance permits the drug's use in treating a broader group of individuals suffering from prostate cancer, specifically those whose cancer has progressed despite other treatments and who possess particular genetic mutations.
According to Seeking Alpha, this marks a significant development, as it is the first new therapeutic option for this specific patient demographic in approximately ten years. The expanded approval targets adult patients with hormone receptor-positive, HER2-negative locally advanced or metastatic breast cancer, and adult patients with prostate cancer that is resistant to castration and has specific genetic alterations.
Truqap is an oral AKT inhibitor, a type of medication that works by blocking a protein called AKT which can contribute to cancer growth. The FDA initially approved Truqap in November 2023 for use in combination with fulvestrant for certain types of breast cancer. The current expansion broadens its applicability to prostate cancer patients who have experienced disease progression following at least one prior hormonal therapy and at least one chemotherapy regimen, and whose tumors exhibit one of the PIK3CA, AKT1, or PTEN gene mutations.
Prostate cancer remains a prevalent and challenging disease, particularly when it becomes resistant to castration and metastasizes. The availability of new treatment modalities is crucial for patients who have exhausted other options. This expanded indication provides a new avenue for medical professionals to consider for a subset of patients with specific biomarkers.
AstraZeneca continues to invest in oncology research, with a focus on developing targeted therapies that address the underlying genetic drivers of cancer. The successful expansion of Truqap's label underscores the potential of precision medicine in improving patient outcomes. Further trials and real-world data collection will be essential to fully understand the long-term impact and benefits of this expanded approval across the eligible patient population.
